NeuraminidasesareusedtocleaveterminalN-acetylneuraminicacid(sialicacid)fromavarietyofglycoproteins.TheenzymefromClostridiumperfringenscleavesterminalsialicacidresidueswhichareα-2,3-α-2,6-orα-2,8-linkedtoGal,GlcNac,GalNAc,AcNeu,GlcNeu,oligosaccharides,glycolipidsorglycoproteins.Therelativerateofcleavagedecreasesintheorder:α-2-3>α-2-6.α-2-8.NeuraminidasefromC.perfringenscleavesα-2-3linkedsialicacidresiduesmostefficiently,comparedtoA.ureafaciens,(SigmaN3642)whichpreferentiallycleavesα-2-6linkedresidues.
Theuseofneuraminidasetoremovesialicacidresiduesfromglycoproteinsoncellsurfaceshasbeenfrequentlyreported.Generally,procedureshaveindicatedusingneuraminidaseinPBSat37°Cfor30minutes,followedbyseveralwashingswithPBS.Treatmentoftissuesectionswithneuraminidaseatmuchlowerconcentrationsrequirelongerincubation:for1-4U/mLin0.1MacetatebufferpH4.2-5,from2to20hoursat37°C.
