Alcoholdehydrogenases(ADH)areafamilyofenzymesthatcatalyzestheconversionofalcoholstoaldehydes,withtheconcomitantreductionofNAD+toNADH.Inhumans,therearenineisozymesofADH,withthemajorityofADHactivityoccurringintheliver.ADHfamilymembersaretheprimaryenzymesinvolvedinalcoholdetoxification.GeneticvariationsinADHenzymesresultindifferencesinADHactivityandtolerancesforalcohol,andmayregulatesusceptibilitytoalcoholism.
TheAlcoholDehydrogenaseActivityAssaykitprovidesasimpleanddirectprocedureformeasuringADHactivityinavarietyofsamples.ADHactivityisdeterminedusingisopropanolasthesubstrateinanenzymereaction,whichresultsinacolorimetric(450nm)productproportionaltotheenzymaticactivitypresent.OneunitofADHistheamountofenzymethatwillgenerate1.0μmoleofNADHperminuteatpH8.0at37°C.
