Unlikemanyotherpeptidases,pepsinhydrolyzesonlypeptidebonds,notamideoresterlinkages.Thecleavagespecificityincludespeptideswithanaromaticacidoneithersideofthepeptidebond,especiallyiftheotherresidueisalsoanaromaticoradicarboxylicaminoacid.Increasedsusceptibilitytohydrolysisoccursifthereisasulfur-containingaminoacidclosetothepeptidebond,whichhasanaromaticaminoacid.Pepsinwillalsopreferentiallycleaveatthecarboxylsideofphenylalanineandleucine,andtoalesserextentatthecarboxylsideofglutamicacidresidues.Itdoesnotcleaveatvaline,alanine,orglycinelinkages.Z-L-tyrosyl-L-phenylalanine,Z-L-glutamyl-L-tyrosine,orZ-L-methionyl-L-tyrosinemaybeusedassubstratesforpepsindigestion.Pepsinisinhibitedbyseveralphenylalanine-containingpeptides.
