Allcollagenmoleculesarecomposedofthreepolypeptidechainsarrangedinatriplehelicalconformation,withaprimarystructurethatismostlyarepeatingmotifwithglycineineverythirdpositionandprolineor4-hydroxyprolinefrequentlyprecedingtheglycineresidue.TypeIVcollagenoccursonlyinthebasementmembranesandcontainsuptosixgeneticallydistincta-chains.
Application
TypeIVcollagenhasbeenfoundtoplayakeyroleinangiogenesis,neurologicaldiseasesandmetastasis.Collagenfromhumanplacentahasbeenusedtoassessthebioelectriceffectsofquinineonairwayepithelialcells,tostudytheselectivetoxicityofengineeredlentviruslyticpeptidesandinaparticleaggregationassayfortherapiddetectionoffibronectin,fibrinogenandcollagenreceptorsonStaphylococcusaureus.Ithasalsobeenusedinamagnesium-dependent,collagen-bindingassayduringcharacterizationofhumanlungtumor-associatedantigens.

Thisproductissuppliedasasolutionin0.5Maceticacid.Itcanbeusedtocoattissue-culturesurfacestopromotecellattachment.Theamountusedintheseapplicationsisdependentonthecelltype,plasticwareandincubationtime.Arecommendedamountis0.5-5μg/cm2.Itwaspreparedbypepsinextractionandchromatographicpurification.
